Proline, glutamate and leucine rich protein 1 (PELP1), also known as modulator of nongenomic actions of the estrogen receptor (MNAR), is a scaffold protein, encoded by the gene mapped to human chromosome 17p13.2. The encoded protein is composed of 1130 amino acids and is ubiquitously expressed, but at high levels in the brain, testes, ovaries and uterus. PELP1 is characterized with ten nuclear receptor (NR)-interacting boxes (LXXLL motifs), a zinc finger, a glutamic acid-rich domain and two proline-rich domains.

The proto-oncogene proline, glutamate and leucine rich protein 1 (PELP1) plays a vital role in cancer progression by stimulating several pathways such as cell cycle, ribosome biogenesis, chromatin modifications, DNA damage response, histone methylation and RNA splicing. It acts as a coactivator for several transcription factors, such as estrogen receptor (ESR), ESR2, androgen receptor (AR), E2 factor and signal transducer and activator of transcription 3 (STAT3). PELP1 expression has been observed in various types of human cancers including breast, endometrial, ovarian, salivary, prostate, lung, pancreas and colon.
