Poly(A) binding protein nuclear (PABPN1) is a ubiquitously expressed nuclear protein that binds specifically and with a high affinity to poly(A) tails at the 3′ ends of mRNAs. PABPN1 contains a single RNP domain that separates a glutamate-rich N-terminal domain from an arginine rich C-terminal domain. PABPN1 is encoded by the gene mapped to human chromosome 14q11.2.

synthetic peptide corresponding to a sequence at the C- terminal region of human PABPN1, conjugated to KLH. The corresponding sequence is identical in rat and mouse.

应用

Anti-PABPN1 antibody produced in rabbit has been used in several immunochemical techniques including immunoblotting (~ 55 kDa) and immunofluorescence.

生化/生理作用

Poly(A) binding protein nuclear (PABPN1) plays a vital role in progressive and efficient polymerization of poly(A) tails on the 3′ ends of eukaryotic genes and controls the size of the poly(A) tail to about 250 nt. The ribonucleoprotein (RNP) domain and the C-terminal domain are required for binding to poly(A), and the N-terminus is essential for the stimulation of polyadenylation. Abnormal expansion of a (GCG)n trinucleotide repeat in the coding region of PABPN1 gene leads to autosomal dominant oculopharyngeal muscular dystrophy (OPMD) disease.
