一般描述
Bridging integrator 1 (BIN1) is mapped to human chromosome 2q14.3 and comprises 20 exons. The gene encodes multiple splice variants as close to 10 isoforms. Isoform nine and 10 are ubiquitous, eight is expressed in muscle. Rest other isoforms are expressed in brain. BIN1 comprises N-terminal BAR (Bin1/Amphiphysin/RVS167) domain and C-terminal SRC homology 3 (SH3) domain. It belongs to BAR superfamily and is a membrane deforming protein.

生化/生理作用
Bridging integrator 1 (BIN1) mediates the endosomal vesicle sorting. It also modulates β-site amyloid precursor protein cleaving enzyme 1 (BACE1) and regulates amyloid Aβ production. BIN1 interacts with actin through the BAR domain and also regulates tubulation of T?tubules and clathrin?mediated endocytosis. Single nucleotide polymorphisms in BIN1 and its altered expression is implicated in the pathogenesis of Alzheimer′s disease. Mutations in the BAR domain of the BIN1 leads impairment in T-tubule organization in centronuclear myopathy (CNM).
