一般描述

Mannitol dehydrogenase (MDH) is a NAD-dependent enzyme, obtained from several organisms like,Lactobacillus brevis,Leuconostoc mesenteroides,Absidia glaucaandRhodococcus. MDH obtained fromL. mesenteroidesexhibits similarity in sequence with the medium-chain dehydrogenase/reductase protein family. It also contains four identical subunits of about 38 kDa in size.
The molecular weight of mannitol dehydrogenase is 136 kDa.
Mannitol dehydrogenase is a cytoplasmic enzyme whose primary role is the regulation of intracellular levels of the sugar alcohol mannitol in plants. It catalyzes the pyridine nucleotide dependent reduction of fructose to mannitol.

应用

It has been used in a study to assess glucosylglycerol and glucosylglycerate as enzyme stabilizers.
Can be used for the production of D-mannitol.
This preparation is useful in the determination of mannitol in urine.

Mannitol dehydrogenase (MDH) helps to catalyze the pyridine nucleotide-dependent oxidation of D -mannitol to D -fructose. Mannitol dehydrogenase (MDH) obtained fromL. mesenteroidesis highly specific for the conversion fructose and mannitol.

单位定义

在 pH 5.3 和 30℃ 且存在 NADH 的条件下，一单位本品每分钟将还原 1.0μmol D -果糖。如上定义的一单位酶相当于如 Lunn 等人所述的约 1.8 单位酶，并且在 pH 8.6 和 40℃ 且存在 NAD 的条件下，一单位酶每分钟将氧化 1.0μmol D -甘露醇。
