Description
MDA-MB-231来自患有转移乳腺腺癌的51岁白种人女病人的胸水。在裸鼠和ALS处理的BALB/c小鼠中,它能形成低分化腺癌(III级)。细胞表达WNT7B致癌基因。此细胞株可能作为转染宿主。我库冻存时为P41代。每个批次均通过本库支原体检测,结果阴性。经本库STR检测无误,STR鉴定结果为:Amelogenin: X;CSF1PO: 12,13;D13S317: 13;D16S539: 12;D5S818: 12;D7S820: 8,9;THO1: 7,9.3;TPOX: 8,9;vWA: 15,18。

人乳腺癌细胞MDA-MB-231完全培养液配方(100 ml):
L-15 (Hyclone, SH30525.01  或Invitrogen 11415-064)90 ml
FBS (Gibco)         10 ml
气相:空气,100%。温度:37摄氏度。
参考传代比例:1:3
参考传代周期:4-6天
参考换液频率:每周2次
冻存液配方:完全培养液95%,DMSO 5%
【注意事项:】
1.该细胞使用的L-15基础培养液由磷酸盐和游离碱氨基酸代替了碳酸氢钠缓冲体系。   该培养基必须在没有CO2平衡的环境中培养细胞。不能在普通的5% CO2细胞培养箱中培养,培养液颜色会明显变黄。
2.细胞形态大部分为纺锤状,也有部分细胞呈圆形,存在少量悬浮细胞。
3.如果发现细胞生长极其缓慢,可提高血清含量至20%。
4. 我库冻存时,每支冻存管约含7×105细胞量,体积为500μl,预期存活率70%,建议复苏至1个T25培养瓶中。
